My scientific interest is in the general area of the mechani stic enzymology of redox enzymes, with a specific interest in flavin-depe ndent enzymes. The long-term objectives are to understand how enzymes can influence the energetics of reaction intermediates and transition states . These aspects are being studied by steady state kinetics and rapid reac tion techniques using pH, viscosity and kinetic isotope effects, as well as mutagenesis and X-ray crystallography. Several enzymes from diverse ce llular sources are currently investigated, including choline oxidase, nit ronate monooxygenase, glycolate oxidase, D-arginine dehydrogenase, and ot hers. Significant advances on the understanding of the enzyme-cvatalyzed, flavin-dependent mechanisms for alcohol and amine oxidation have been ob tained. Results from my laboratory aided in the reclassification of 2-nit ropropane dioxygenase as nitronate monooxygenase by the IUBMB in 2010.
