Iron-sulfur (Fe-S) cluster-bound transcription factors play vital roles in all forms of life. This vividly contrasts with the scarcity of information on their structural and molecular mechanism, as exemplified by the WhiB-like (Wbl) proteins. The Wbl proteins are a unique group of [4Fe-4S] cluster-bound proteins in Actinobacteria and highly conserved in mycobacteria, including the notoriously persistent pathogen Mycobacterium tuberculosis (Mtb).
