Methods for Fast and Inducible Bioorthogonal Chemistry

This seminar will discuss recent advances in development of the tetrazine ligation– the fastest known bioorthogonal reaction. Tetrazine ligation has been used broadly by the scientific community, and finds application across chemical biology, medicine, nuclear medicine, and material science. Our group has also developed the only general method for the synthesis of trans-cyclooctenes, molecules of high importance to the field of bioorthogonal chemistry. Using this method as a key step, we have synthesized new trans-cyclooctene analogs with exceptionally fast kinetics. Also described will be new tools for the safe synthesis and direct introduction of ‘minimalist’ tetrazines to molecules including API’s and fluorescent reporters. Collaboratively, has used bioorthogonal chemistry in conjunction with new reactivity for detecting oxidative stress in living cells, and our group has used the interfacial nature of tetrazine ligation as a new way to create and pattern biomaterials. Finally, the talk will discuss the develop efficient catalytic methods for ‘turning on’ rapid bioorthogonal chemistry in cellular context. Discussed will be new tool molecules with high stability in the cellular environment in their ‘off’ state, and the fastest bioorthogonal reactions to date in their ‘on’ state. The methodology serves as a new platform for rapidly inducing and tracking fluorescence in drug targets at the subcellular level, and as a new method of drug release where localized photocatalysts are used to drive the local release of a cytotoxic reagent from a photocaged prodrug.
Joseph M. Fox is Professor of Chemistry in the Department of Chemistry and Biochemistry at the University of Delaware, where he also is the Director of the NIH-funded Center of Biomedical Research Excellence on Molecular Discovery.
